Jacksonville is Florida’s largest city by land area, and it sits about 375 miles east of Destin — roughly a 5-hour drive depending on traffic through Tallahassee. It’s a straightforward interstate route that thousands of Floridians make every summer, and with a little planning it’s a genuinely pleasant road trip.

There’s really only one sensible route: take I-10 West from Jacksonville all the way through Tallahassee, then drop south on US-331 (or US-85 if you prefer) into the Destin–Fort Walton Beach corridor. The total distance is about 375 miles and Google Maps will clock it at 4h 45m to 5h 15m in normal conditions.
Alternate: Some people take US-90 (the old pre-interstate highway) instead of I-10. It’s slower but runs through Quincy, Marianna, and other small towns if you want a more scenic drive. Add 45–60 minutes.
